  • What are the main claims of the paper and how important are they?

The author has attempted to write a review on how oxidative stress mediated by thyroid hormones can be protected-but it doesnr make any sense as first of all in rats where they have conducted all there experimentation the effect of thyroid gland is in prepubertal rats only in affecting sertoli cell proliferation and thereby their maturation as well and affect by oxidative stress increase or decrease by hyper or hypo and in humans  b3esides that there is no proven role and umpteen articles are there on role of oxidative stress in male fertility and author has not even mentioned prepubertaL in the studies they have used to quote and roles of alpha tocopherol(vit E,VIT C as natural anti oxidants is well known and thyroid hormone has multiple role besides oxidative stress ,in affecting testiculat development and function-so the idea of this review doesnt make much sense from any therapeutic benefit or otherwise.

  • Are these claims novel? If not, please specify papers that weaken the claims to the originality of this one.

No but role of thyroid hotmone mediated oxidative stress doesnt make any sense-in man thyroid has a big physiological rile in metabolism and is required for developmwent, metabolism, and the oxidative stress authors have found is artificially ctreated in animal models by giving PTU,Methimazole or T3/T4 and in humans no proven role of any such hyper/hypothyroidism related oxidative stress has been documented although in animal models other important role of thyroid in development of testis has been documented and umpteen articles on oxidative stress and infertility with roles of vitE,vit c AND OTHER anti oxidants like luycopene atc with no statistical proven benefit in fertility has been shown despite theoretical advantages 1)tremellen K Oxidative stress and male infertility.Hum Reprod Update 2008;14(3):243-258.2)Aggarwal A,Sharma RK,Nallela KP,ThomasAJ,AlvarezAJ,Sikka SC.Reactive oxygen species as an independent marker of male factor infertility.Fert Steril 2006;86:878-885.3)Wagner MS,Wajner MS,Maia LA.The role of thyroid hormone in testicular development and function.J of Endocrinol 2008;199:351-365.4)Zamoner A,Barreto PK,Filho WD,Sell F,Woehl M,Rodrigues Guma FC,Pessoa -Pureur R,Silva MB.Propyl thiouracil-induced congenital hypothyrooidism upregulates vimentin phoisphorylation and depletes antioxidanr defense in immature rat testes.J of Mol Endocrinol 2008;40:125-135.-this is similar to the prepubertal experiments done by author and adds to the mechanism of depleting antioxidant defenses

  • Are the claims properly placed in the context of the previous literature?

    Title is misleading-orherwise ok

  • Do the results support the claims? If not, what other evidence is required?

    NA

  • If a protocol is provided, for example for a randomized controlled trial, are there any important deviations from it? If so, have the authors explained adequately why the deviations occurred?

    NA

  • Is the methodology valid? Does the paper offer enough details of its methodology that its experiments or its analyses could be reproduced?

    NA

  • Would any other experiments or additional information improve the paper? How much better would the paper be if this extra work was done, and how difficult would such work be to do, or to provide?

    NA

  • Is this paper outstanding in its discipline? (For example, would you like to see this work presented in a seminar at your hospital or university? Do you feel these results need to be incorporated in your next general lecture on the subject?) If yes, what makes it outstanding? If not, why not?

No-Rather doesnt make any sense from any therapeutic value for mankind

  • Other Comments:

If at all to give it any signoficance the author should combine his findings and update the literature till date on role of thyroid gland in the development of sertoli cell ,leydig cell metsabolism,AMH,T hyroid receptor alpha 1 or 2 influence ,absence of beta rather than just giving an update of tole of thyroid on oxidative stress as all animal studies show not much role beyond puberty in testis development of thryroid band highlight the importance of their weork on apoptosis caused by hypo/hyper thyroidism at pubertal age.-summarixe effects on AMH,aR etc and connexin43 and other updatedroles of thyroid in testicular sertoli cell proliferation and maturation and further affect germ cwll development.

I am very much thankful to Prof. Kaur for critically evaluating the paper “Sahoo DK. Testicular Protection From Thyroid Hormone Mediated Oxidative Stress. WebmedCentral REPRODUCTION 2013;4(5):WMC004252” I understand that the information given in the review is mainly studied in animal systems i.e. in rat (a mammalian model). Hence, further extensive studies are needed particularly for its application in human therapy.

Review of the article

  • What are the main claims of the paper and how important are they?

This paper is a review article on general antioxidant defence status of testis and how it is disturbed by hypo-and hyperthyroidism and is there any ways to protect testis from such type of oxidative stress.

  • Are these claims novel? If not, please specify papers that weaken the claims to the originality of this one.

Yes. This is a review article. The author tries to make a story about the general antioxidant defence status of testis and how it gets disturbed by hypo-and hyperthyroid conditions and the ways to protect testis from such type of oxidative stress.

  • Are the claims properly placed in the context of the previous literature?

Yes. The author cited his work along with others. I found Sahoo et al., 2005 is missing from the reference section.

  • Do the results support the claims? If not, what other evidence is required?

    Yes

  • If a protocol is provided, for example for a randomized controlled trial, are there any important deviations from it? If so, have the authors explained adequately why the deviations occurred?

    NA

  • Is the methodology valid? Does the paper offer enough details of its methodology that its experiments or its analyses could be reproduced?

    NA

  • Would any other experiments or additional information improve the paper? How much better would the paper be if this extra work was done, and how difficult would such work be to do, or to provide?

Sometimes, it looks in the manuscript the author tried to focus his findings only. Studies on different ways to protect hypothyroid rat testis from oxidative damage would be great, however due to lack of any literature the author only focused on T3 treatment and withdrawl PTU effects (transient hypothyroid condition).

  • Is this paper outstanding in its discipline? (For example, would you like to see this work presented in a seminar at your hospital or university? Do you feel these results need to be incorporated in your next general lecture on the subject?) If yes, what makes it outstanding? If not, why not?

    Yes.

Yes, this paper seems like a report on a new condition but only it is not new. also, seems like a report for jurnals or websites but not for clinical databases, since most of the references wrote the same but with trials or reporting cases or as areview with meta-analysis but not stating facts collectivly which is better in a book chapter.

  • Is this paper outstanding in its discipline? (For example, would you like to see this work presented in a seminar at your hospital or university? Do you feel these results need to be incorporated in your next general lecture on the subject?) If yes, what makes it outstanding? If not, why not?

No, all references wrote on the same subject and in pubmed 1800 other reference wrote on the subject.

I am very much thankful to Dr. Othman for critically evaluating the paper “Sahoo DK. Testicular Protection From Thyroid Hormone Mediated Oxidative Stress. WebmedCentral REPRODUCTION 2013;4(5):WMC004252”. I would like to state that this is a small review article focusing on the available information on studies on testicular protection related to hypothyroid and hypothyroid induced oxidative stress. I understand that there are number of papers available related to oxidative stress in different tissues and conditions, but this paper is focused only on testicular protection related to hypothyroid and hyperthyroid induced oxidative stress.
