Research project: Designing a new recipe for cell-protein interactions.
Cell-surface interactions are fundamental to in vivo implant biology, biosensor, cell microarray and performing cell biology to chip format. Controlling material surface features at nanoscale length to optimize cell response is instrumental to success of biomedical device/implant. Using pulsed microplasma cluster source (PMCS) equipped with supersonic cluster beam (SCBD) which is a home built facility at our lab; we developed novel protocols for micro-nanostructuring of solid state materials (TiOx/ZrOx/CNx) in chip format. The novelty of the technique lies in its facility of decoupling nanoscale topography with surface chemistry and vice versa to study the influence of surface physico-chemical properties on the performance of the materials in contact with complex biological environment. The most significant step of my PhD dissertation is the development of high throughput approach for the nanostructured thin film synthesis and characterization. We developed recipes for novel solid state material library design with gradient in nanoscale morphology roughness and wettability, based on beam engineering of PMCS/SCBD apparatus. Further, using multiplex fluorescence microscopy, probed the “hit spots” for optimum material surface-protein-cell interactions using prokaryotic and eukaryotic (mammalian and bacterial) cells with high content image analysis software (ImageJ, IMARIS 7.0, Bitplane's core system). On single material library with more than 1000 gradient spot array, it is possible to predict the optimum material surface properties to promote mammalian cell-interaction and/or inhibit bacterial colonization and vice versa. Technique routinely used so far polymeric material design, proposes new avenues for solid state materials development and discovery.

Material Fabrication: Colloidal synthesis and characterization of metal nanoparticles via chemical and green (microbial/plant) routes
Operation and handling of pulsed microplasma cluster source (PMCS) for supersonic cluster beam deposition (SCBD) of nanostructured thin film of transition metal oxides (titanium, zirconium, carbon). Atomic force microscopy (AFM) for nanoscale morphology characterizations, Optical prifilometry analysis, and water contact angle (WCA) measurement.
Developed high throughput (HTP) approaches for synthesis and characterization of material gradient library of TiOx/ZrOx/CNx and nanostructured surface-protein-surface interactions on single chip. High content optical profilometric analysis using Apex 2DTM  software for nanoscale step thickness, rms roughness and surface skewness analysis.
Biological Assays: Nanoparticle-cell interactions assays for cytotoxicity evaluation (cell culture: PC12, MG63, SHSY5Y, HeLa, MDCK, U2OS cell lines.
Molecular biology and Microscopy techniques: MTT assay, protein extraction purification, SDS-PAGE for protein identifications bright filed/confocal scanning laser microscopy (CLSM).
Designed and developed micro-nanostructured platform to control cell-surface interaction on chip for cell assays and implants surface optimization. During training, pursued different polymer spin coating, lithography, routine mammalian cell culture protocol and viability assays (Annxin V/MTT), immunostaning, CLSM. Multi content fluorescent microscopy with full automations.
