  • What are the main claims of the paper and how important are they?

    The paper claims that resveratrol, as well as other dietary constituents, create cytotoxicity in cells that have damaged DNA repair pathways. This is important because the study conducted with resveratrol in cells with DNA repair deficient pathways support the claim made with suffcient data and suggest why other dietary constituents, previously thought to be safe, could also be causing adverse effects.


  • Are these claims novel? If not, please specify papers that weaken the claims to the originality of this one.

    Yes. Although it has been previously identified that resveratrol can cause genotoxic activity, this paper describes how the cytotoxicity is most evident in cells lacking DNA repair mechanisms, not all cells.


  • Are the claims properly placed in the context of the previous literature?

    Yes.


  • Do the results support the claims? If not, what other evidence is required?

    Yes.


  • If a protocol is provided, for example for a randomized controlled trial, are there any important deviations from it? If so, have the authors explained adequately why the deviations occurred?

    Yes. MTT cell proliferation assay.


  • Is the methodology valid? Does the paper offer enough details of its methodology that its experiments or its analyses could be reproduced?

    The methodology is valid and the experiments could be effectively reproduced. However, it seems that additional studies should be completed.


  • Would any other experiments or additional information improve the paper? How much better would the paper be if this extra work was done, and how difficult would such work be to do, or to provide?

    This paper could be improved by further explaining the data found with the MTT cell proliferation assay rather than just referencing the data collected. It would not take very much to add information about specific cell lines and possible reasons why certain cell lines behaved differently than other cell lines. For example, why did NMR and NHEJ deficient pathways show the least amount of change compared to parent cell lines.

    Yes. Evidence of why resveratrol might be cytotoxic in some instances.


  • Other Comments:

    This article was very informational and well written. The Introduction was thorough enough that the results of the study could be understood without having extensive knowledge in the reserach area. However, the Resutls and Discussion section could be expanded upon more.

    The main claim is the cytotoxic effect of reservetorl which is mediated through ROS mediated DNA damage.


  • Are these claims novel? If not, please specify papers that weaken the claims to the originality of this one.

    Moderetely novel


  • Are the claims properly placed in the context of the previous literature?

    The claims were partially addresed


  • Do the results support the claims? If not, what other evidence is required?

    The authors had performed only MTT assay to substanciate their claim.


  • If a protocol is provided, for example for a randomized controlled trial, are there any important deviations from it? If so, have the authors explained adequately why the deviations occurred?

    NA


  • Is the methodology valid? Does the paper offer enough details of its methodology that its experiments or its analyses could be reproduced?

    Additional functional and biochemical assays are required


  • Would any other experiments or additional information improve the paper? How much better would the paper be if this extra work was done, and how difficult would such work be to do, or to provide?

    As mentioned previously mechanism has to be explored.

    The mecahnsim of reservetorl and its action on DNA damage is well known.
