  • What are the main claims of the paper and how important are they?

The main claim of this article is to formulate effective fast dissolving tablet of gliclazide to improve patient compliance to pediatric or geriatric population. It is important because these patients have difficulties while swallowing drugs so this formulation will make them easier to take the drug which will dissolve in the mouth.

  • Are these claims novel? If not, please specify papers that weaken the claims to the originality of this one.

Yes, unless the drug metabolizes thoroughly in the mouth before it gets to its target.

  • Are the claims properly placed in the context of the previous literature?

Yes, the claims are appropriately placed in the context of the previous literature.

  • Do the results support the claims? If not, what other evidence is required?

Yes, there are nine tables and three figures which are enough to support the claims.

  • If a protocol is provided, for example for a randomized controlled trial, are there any important deviations from it? If so, have the authors explained adequately why the deviations occurred?

No, there are no important deviations in the article.

  • Is the methodology valid? Does the paper offer enough details of its methodology that its experiments or its analyses could be reproduced?

It is valid and reproducible if the person is in the same field.

  • Would any other experiments or additional information improve the paper? How much better would the paper be if this extra work was done, and how difficult would such work be to do, or to provide?

In vivo or clinical PK data may improve the paper and confirm the effect of combination of superdisintegrants. However, it is complicated. It will cost more money, time, and effort. The official requirements, what the article did, would be enough because they were already approved methods.

  • Is this paper outstanding in its discipline? (For example, would you like to see this work presented in a seminar at your hospital or university? Do you feel these results need to be incorporated in your next general lecture on the subject?) If yes, what makes it outstanding? If not, why not?

Yes, it is a quite outstanding and well organized paper. I will use this paper as a example of a well performed article in the lecture or seminar. Because the article successfully formulate fast dissolving tablet and it explained clearly for the justification and process of their research.

  • What are the main claims of the paper and how important are they?

The article claims that the research formulated directly compressible fast dissolving tablets of Gliclazide. They claimed their C5 batch was optimized and disintegrates in 24.7 seconds. The claims are important because the faster the disintegrant the easier and better the delivery for patients.

  • Are these claims novel? If not, please specify papers that weaken the claims to the originality of this one.

The claims are novel in that they combine different superdisintegrants in a way that has not been tested. A review paper by Mohanachandran, Sindhumol and Kiran in 2011, looks at some of these superdisintegrants. However, they do not combine them, but state that they could be combined. The originality is there, it is just taking something known about and combining them and testing them in new ways. Panigrahi, Chowdary, Mishra, Bhowmik, and Beis article which did another study on combinations of superdisintegrants. However, the particual drug was different and the exact combination was different, they looked at croscarmellose sodium, crospovidone, and sodium starch glycolate just like they did here. Both drugs are used in diabetes.

  • Are the claims properly placed in the context of the previous literature?

Yes, the 2011 (Mohanachandran, Sindhumol and Kiran) review looks at crospovidine and others in 2011. Fast dissolving tablets with combination of superdisintegrants were looked at by the same group with a different drug at the same time. This means the literature matches the types of studies being done in this article.

  • Do the results support the claims? If not, what other evidence is required?

Yes the data does support the claims. However, it would have been better to specifically state more of the data within the text for easy analysis.

  • If a protocol is provided, for example for a randomized controlled trial, are there any important deviations from it? If so, have the authors explained adequately why the deviations occurred?

    No

  • Is the methodology valid? Does the paper offer enough details of its methodology that its experiments or its analyses could be reproduced?

The materials and methods sections were short. When looking at another paper by them, they were more explicit. I would have liked to have seen more discriptions in the text about the methodology. However, it is valid and probably more easy to reproduce by someone who has worked with them.

  • Would any other experiments or additional information improve the paper? How much better would the paper be if this extra work was done, and how difficult would such work be to do, or to provide?

I don't think so, they make the points they want to make. The article could have been worded in a more organized way leading the reader through the paper better, as already stated.

  • Is this paper outstanding in its discipline? (For example, would you like to see this work presented in a seminar at your hospital or university? Do you feel these results need to be incorporated in your next general lecture on the subject?) If yes, what makes it outstanding? If not, why not?

Yes, it is outstanding that they found a formulation that performed the way they needed it to, to all the specifications needed. In research, that doesn't always happen.

8 Is the quality of the diction satisfactory? No
9 Are the illustrations and tables necessary and acceptable? No
10 Are the references adequate and are they all necessary? Yes
11 Are the keywords and abstract or summary informative? Yes
  • Other Comments:

The original research paper on different formulations of the drug gliclazide showed that there are two formulations that have a good release profile for a fast-dissolving tablet over 30 minutes. However, the authors mentioned in the conclusions that formulations C5 and C8 presented with the best dissolution profile but then referred to preparations C4 and C8 in all of the graphs. This may be a simple labeling error but should definitely be corrected. Furthermore, the authors should carefully read over the manuscript and make any necessary diction corrections.

In the present investigationj use of combination superdisintegrants for better release rate of drug from fast disolving tablets of Gliclazide is one of the novel approach done so far.

As Glicalazide is poorly soluble drug it needs enhanced solubility in body fluid (in vivo), hence the approach decided by the author is one of the way to enhance the solubility and improvment of drug dissolution.

Here the author suggested to use rational combinations of disintegrants in appropriate proportions  for futher studies.
